Applications of BBa_K2179003

This part works extremely well when cloned into the DNA 2.0 PCB-38-441 backbone using BsaI. In this context, the gene is under tight LacI regulation consisting of a plasmid borne gene for the LacI repressor and two Lac operators O1 and O2 that flank the promoter. To generate oxygen from chlorite, cell are grown to an OD600 of 0.5. Folowing the addition of IPTG (1.0mM final), levulinic acid (2.0mM final) and FeSO4 (2mM) final cell are incubated at 37 degrees C over night. The addition of chlorite (0.5M final) produce the immediate bubbling of the culture as a result of oxygen production.
